Function in Host |
Functions both as NADH oxidoreductase and as regulator ofapoptosis. Inresponse to apoptotic stimuli, it is released from the mitochondrionintermembrane space into the cytosol and to the nucleus, where itfunctions as a proapoptotic factor in a caspase-independent pathway. The soluble form (AIFsol) found in the nucleus induces 'parthanatos'i. e. caspase-independent fragmentation of chromosomal DNA. Binds to DNA in a sequence-independent manner. Interacts with EIF3G, and thereby inhibits the EIF3machinery and protein synthesis, and activates caspase-7 to amplifyapoptosis. Plays a critical role in caspase-independent, pyknotic cell death in hydrogen peroxide-exposed cells. In contrast, participates in normal mitochondrialmetabolism. Plays an important role in the regulation of respiratorychain biogenesis by interacting with CHCHD4 and controlling CHCHD4mitochondrial import.

Method Description | To detect the role of host protein AIFM1 in viral infection, AIFM1 protein knockout Calu-3 Cells were infected with SARS-COV-2 for 7 - 9 Days , and the effects on infection was detected through CRISPR-based genome-wide gene-knockout screen. | ||||||||
Results | It is reported that knockout of AIFM1 increases SARS-CoV-2 RNA levels compared with control group. |
