Binds double-stranded DNA. Binds preferentially tosupercoiled DNA and cruciform DNA structures. Seems to be involved intranscriptional regulation. May function as a transcriptionalrepressor. Could have a role in the regulation of hematopoieticdifferentiation through activation of unknown target genes. Controlscellular proliferation by modulating the functions of cell cycleregulatory factors including p53/TP53 and the retinoblastoma protein. May be involved in TP53-mediated transcriptional activation byenhancing TP53 sequence-specific DNA binding and modulating TP53phosphorylation status. Seems to be involved in energy-level-dependentactivation of the ATM/ AMPK/TP53 pathway coupled to regulation ofautophagy. May be involved in regulation of TP53-mediated cell deathalso involving BRCA1. May be involved in the senescence of prostateepithelial cells. Involved in innate immune response by recognizingviral dsDNA in the cytosol and probably in the nucleus. After bindingto viral DNA in the cytoplasm recruits TMEM173/STING and mediates theinduction of IFN-beta. Has anti-inflammatory activity and inhibits theactivation of the AIM2 inflammasome, probably via association withAIM2. Proposed to bind viral DNA in the nucleus, such as of Kaposi'ssarcoma-associated herpesvirus, and to induce the formation of nuclearcaspase-1-activating inflammasome formation via association withPYCARD. Inhibits replication of herpesviruses such as humancytomegalovirus (HCMV) probably by interfering with promoterrecruitment of members of the Sp1 family of transcription factors. Necessary to activate the IRF3 signaling cascade during human herpessimplex virus 1 (HHV-1) infection and promotes the assembly ofheterochromatin on herpesviral DNA and inhibition of viral immediate-early gene expression and replication. Involved in the MTA1-mediatedepigenetic regulation of ESR1 expression in breast cancer.

Method Description | To detect the role of host protein IFI16 in viral infection, IFI16 protein knockout Calu-3 Cells were infected with SARS-COV-2 for 7 - 9 Days , and the effects on infection was detected through CRISPR-based genome-wide gene-knockout screen. | ||||||||
