Scaffold protein of the presynaptic cytomatrix at the activezone (CAZ) which is the place in the synapse where neurotransmitter isreleased. After synthesis, participates in theformation of Golgi-derived membranous organelles termed Piccolo-Bassoontransport vesicles (PTVs) that are transported along axons to sites ofnascent synaptic contacts. At the presynaptic activezone, regulates the spatial organization of synaptic vesicle cluster, the protein complexes that execute membrane fusion and compensatoryendocytosis. Organizes as well the readily releasablepool of synaptic vesicles and safeguards a fraction of them to be notimmediately available for action potential-induced release. Functions also in processes other than assembly such asthe regulation of specific presynaptic protein ubiquitination byinteracting with SIAH1 or the regulation of presynaptic autophagy. Mediates also synapse to nucleus communication leading toreconfiguration of gene expression by associating with thetranscriptional corepressor CTBP1 and by subsequently reducing the sizeof its pool available for nuclear import.
