Multifunctional RNA-binding protein involved in theregulation of pre-mRNA splicing, mRNA stability and mRNA translationimportant for cell fate decision and differentiation. Plays a major rolein pre-mRNA alternative splicing regulation. Mediates preferentially muscle-specific exoninclusion in numerous mRNAs important for striated cardiac and skeletalmuscle cell differentiation. Binds to intronicsplicing enhancer (ISE) composed of stretches of GU-rich motifslocalized in flanking intron of exon that will be included byalternative splicing. Involved in embryonic stem cell (ESC) transition to cardiac cell differentiation by promoting pre-mRNAalternative splicing events of several pluripotency and/ordifferentiation genes. Plays a role in the regulationof mRNA stability. Binds to 3'-untranslated region (UTR) AU-richelements in target transcripts, such as CDKN1A and MYOG, leading tomaintain their stabilities. Involvedin myogenic differentiation by regulating MYOG levels. Binds to multiple regions in the mRNA 3'-UTR of TP63isoform 2, hence inducing its destabilization. Promotes also the destabilization of the CHRM2 mRNA via its binding toa region in the coding sequence. Plays a role in theregulation of mRNA translation. Mediates repressionof p53/TP53 mRNA translation through its binding to U-rich element inthe 3'-UTR, hence preventing EIF4E from binding to p53/TP53 mRNA andtranslation initiation. Binds to a huge amount ofmRNAs. Required for embryonic heart development, sarcomer and M-band formation in striated muscles. [1-5]

Method DescriptionTo detect the role of host protein RBM24 in viral infection, RBM24 protein knockout Calu-3 Cells were infected with SARS-COV-2 for 7 - 9 Days , and the effects on infection was detected through CRISPR-based genome-wide gene-knockout screen.
ResultsIt is reported that knockout of RBM24 leads to the decreased SARS-CoV-2 RNA levels compared with control group.
